Elute with 40% ethyl acetate
Prepare previously 40% ethyl acetate in naphtha, by mixing 3 parts naphtha with 2 ethyl acetate. Repeat the process as outlined earlier. A very dark green band moves off. Keep going until it becomes lighter in color.
Elute with 60% ethyl acetate
Now elute with 3:2 ethyl acetate:naphtha. A yellow band comes off. Collect this in a separate container. The salvinorin A comes off with this yellow band. Do this about 3 times.
3 fractions
Thew dark green on the left contains the naphtha and 40% ethyl acetate eluants. The light green contains the end of the 40% fraction, when it turned from dark to light green. The one on the right is yellow in color and is the 60% ethyl acetate fraction. 94% of the salvinorin was in this fraction.
